種植于硅橡膠膜上的大鼠纖維環(huán)細(xì)胞的表型特征研究

Phenotypic characteristics of rat annulus fibrosus cells cultured on flexible silicone membranes
【Abstract】 AIM: To compare the phenotypic characteristics of rat annulus fibrosus (AF) cells cultured on flexible silicone membranes with the same cells cultured in plastic plates. METHODS: The morphology of AF cells cultured in different substrates was observed under an inverted phasecontrast microscope and a transmission electron microscope. Proteoglycan was stained with toluidine blue for observing its expression. The contents of collagen typeⅠ, Ⅱand aggrecan mRNA were determined by RTPCR. The expression of integrin β1 was monitored by flow cytometry. Cell adhesion to silicone membranes was also measured. RESULTS: The AF cells cultured in different substrates were morphologically undistinguishable. Toluidine blue staining showed that there was also no difference between AF cells cultured on flexible silicone membranes and in plastic plates. The cells growing in different substrates expressed the same levels of collagen typeⅠ, Ⅱ and aggrecan mRNA and integrin β1. AF cells grew well on silicone membranes. CONCLUSION: AF cells cultured on flexible silicone membranes maintain the stability of phenotype and may be appropriate for further studying the metabolic responses to mechanical stimuli at the cellular level.

【摘要】 目的:比較種植于硅橡膠膜或塑料培養(yǎng)板的大鼠纖維環(huán)細(xì)胞的表型特征. 方法:利用倒置相差顯微鏡及透射電鏡觀察種植于不同基質(zhì)的大鼠纖維環(huán)細(xì)胞形態(tài)的變化;利用甲苯胺藍(lán)染色觀檢測蛋白多糖的表達(dá)情況;通過RTPCR法檢測Ⅰ,Ⅱ型膠原及聚集蛋白聚糖mRNA的表達(dá)情況;采用流式細(xì)胞術(shù)觀察細(xì)胞表面整合素β1的表達(dá)情況;同時(shí)檢測了纖維環(huán)細(xì)胞在硅橡膠膜上的黏附情況. 結(jié)果:種植于不同基質(zhì)的纖維環(huán)細(xì)胞形態(tài)學(xué)觀察無明顯差別;在兩種基質(zhì)上的纖維環(huán)細(xì)胞甲苯胺藍(lán)染色無差異;Ⅰ,Ⅱ型膠原及聚集蛋白聚糖mRNA及細(xì)胞膜整合素β1的表達(dá)亦無明顯差異;纖維環(huán)細(xì)胞可以黏附于硅橡膠膜上生長. 結(jié)論:種植于硅橡膠膜上的纖維環(huán)細(xì)胞表型特征無改變,為進(jìn)一步研究力學(xué)刺激對纖維環(huán)細(xì)胞的影響提供實(shí)驗(yàn)基礎(chǔ).

0引言
下腰痛是一個(gè)重要的公共衛(wèi)生問題,雖然引起下腰痛的原因很多,但是椎間盤退變?nèi)允瞧渲饕蛩兀?-2]. 目前的觀點(diǎn)認(rèn)為,椎間盤退變是一個(gè)由生物化學(xué)和生物力學(xué)因素相互作用的復(fù)雜的病理過程. 由于椎間盤周圍的肌肉、韌帶以及椎間盤內(nèi)的靜水壓等因素,椎間盤始終處于負(fù)荷狀態(tài),尤其是椎間盤纖維環(huán)細(xì)胞總是不斷地受到不同形式的拉伸應(yīng)力. 為了從細(xì)胞水平研究生物力學(xué)因素對細(xì)胞的影響,很多研究者設(shè)計(jì)出不同的實(shí)驗(yàn)裝置,對體外培養(yǎng)細(xì)胞施加不同形式的拉伸應(yīng)力[3-4]. 我們也自行設(shè)計(jì)一套對單層生長細(xì)胞進(jìn)行周期性張應(yīng)變的實(shí)驗(yàn)裝置,為對細(xì)胞施加張應(yīng)力,首先需要將纖維環(huán)細(xì)胞培養(yǎng)于彈性硅橡膠膜上,因此有必要對培養(yǎng)于新的基質(zhì)上的細(xì)胞表型穩(wěn)定性進(jìn)行確認(rèn). 細(xì)胞生長基質(zhì)可影響細(xì)胞外基質(zhì)成分的表達(dá),本實(shí)驗(yàn)我們比較了培養(yǎng)于硅橡膠膜和細(xì)胞培養(yǎng)板上的纖維環(huán)細(xì)胞的細(xì)胞外基質(zhì)成分的表達(dá)情況.
1材料和方法
1.1材料SD大鼠(1月齡)由本院實(shí)驗(yàn)動物中心提供. DMEM培養(yǎng)基和Ⅱ型膠原酶為Gibco公司產(chǎn)品,胎牛血清為PAA公司產(chǎn)品,纖維連接素為Roche公司產(chǎn)品,Hepes為Amresco公司產(chǎn)品,TRIzol試劑為Invitrogen公司產(chǎn)品,一步法RTPCR試劑盒為Qiagen公司產(chǎn)品,F(xiàn)ITC標(biāo)記抗整合素β1單抗為BD公司產(chǎn)品. 硅橡膠膜為Dow Corning公司產(chǎn)品,細(xì)胞培養(yǎng)板為Corn
